Leucine Threshold and mTOR Activation
Muscle protein synthesis is primarily regulated through the mechanistic target of rapamycin (mTOR) signaling pathway. Leucine is the dominant activator of this pathway among the amino acids. Research shows that approximately 2–3g of leucine per meal or serving is required to maximally stimulate MPS in trained individuals. Below this threshold, the anabolic response is blunted regardless of total amino acid intake.

Why BCAAs Are Insufficient
Essential amino acids are the only dietary nitrogen source that directly and independently stimulates MPS. Non-essential amino acids, synthesized internally by the body, do not contribute to this anabolic signaling. Isolated BCAA supplementation (leucine, isoleucine, and valine only) can initiate the MPS signal via leucine but cannot complete the synthesis cascade without the remaining 6 essential amino acids. A complete EAA profile produces a meaningfully greater MPS response than BCAAs alone for this reason.
Lysine as the Rate-Limiting Amino Acid in Indian Diets
Lysine is the first limiting amino acid in wheat, rice, corn, and most cereal grains—the dietary staples of the majority of the Indian population. When lysine is deficient, total protein quality falls below the level required to sustain MPS regardless of how much total protein is consumed. Carnosine is a dipeptide composed of beta-alanine and histidine synthesized in skeletal muscle. It is the primary intramuscular buffer against hydrogen ion accumulation during anaerobic glycolysis—the source of acidosis that limits performance in high-intensity efforts. Histidine availability directly determines the rate of carnosine synthesis. EAA supplementation supporting histidine availability contributes to sustained carnosine levels and improved fatigue resistance in repeated high-intensity training bouts.
